Rationale and design of ApoA-I Event Reducing in Ischemic Syndromes II (AEGIS-II): A phase 3, multicenter, double-blind, randomized, placebo-controlled, parallel-group study to investigate the efficacy and safety of CSL112 in subjects after acute myocardial infarction

C. Michael Gibson et al.

Summary: AEGIS-II is a Phase 3 trial investigating the efficacy and safety of CSL112 compared to placebo among high-risk acute MI participants, aiming to determine whether enhancing cholesterol efflux can reduce the rate of recurrent major adverse cardiovascular events. This study will be the first to formally test this hypothesis.

Exploring the therapeutic potential of the bioinspired reconstituted high density lipoprotein nanostructures

Aditya Gupta et al.

Summary: Lipoproteins, especially HDL, are efficient carriers for cholesterol transportation in the body and show promise for drug delivery due to their antioxidant potential and receptor affinity. rHDL, as an endogenously present vehicle, is capable of targeted delivery for various diseases with the potential for enhanced drug delivery through stimuli sensitivity and pH dependent methods. Further research is needed to explore the full potential of rHDL in clinical applications.

Effect of apoA-I PEGylation on the Biological Fate of Biomimetic High-Density Lipoproteins

Dennis Pedersbaek et al.

Summary: The study found that PEGylation of the b-HDL scaffold had minimal effect on the biological fate of the lipids, with both b-HDL and PEG b-HDL sharing similar biological fates including cellular uptake and tumor accumulation. This highlights the dynamic nature of b-HDL particles, where the biological fate of the components (lipids and scaffold) can differ. A phenomenon that may also apply to other multicomponent nanoparticles.
