A Phase 1b Safety Study of SER-287, a Spore-Based Microbiome Therapeutic, for Active Mild to Moderate Ulcerative Colitis

Matthew R. Henn et al.

Summary: In this study, a phase 1b trial was conducted in 58 adults with active mild-to-moderate UC to evaluate the safety and efficacy of SER-287, an oral formulation of Firmicutes spores. The results showed that patients who received vancomycin preconditioning and daily doses of SER-287 had a higher clinical remission rate at week 8 compared to placebo, indicating that engraftment of dose species was facilitated by vancomycin preconditioning and daily dosing of SER-287.

Hybrid Living Capsules Autonomously Produced by Engineered Bacteria

Daniel P. Birnbaum et al.

Summary: This study presents a simple approach for producing highly programmable bacterial cellulose (BC) materials through incorporating engineered E. coli, expanding the functionality of BC-based living materials. By coculturing Gluconacetobacter hansenii with engineered E. coli in glucose-rich media, robust cellulose capsules are produced, which can sequester specific biomolecules and catalyze reactions in the environment. Additionally, capsules capable of altering their physical properties through enzyme-induced biomineralization are generated.

The evolution of commercial drug delivery technologies

Ava M. Vargason et al.

Summary: This review article discusses how challenges in delivering small molecules, nucleic acids, peptides, proteins, and cells have led to the development of commercial products and are now guiding the delivery of live-cell therapeutics. Drug delivery technologies have enabled the development of pharmaceutical products that enhance therapeutic delivery, minimize off-target effects, and improve patient compliance. Three drug delivery paradigms have been developed to aid in the successful delivery of small molecules and macromolecules, contributing to the initial clinical success of each therapeutic class.
