MS-444, a myosin light chain kinase and HuR inhibitor, is important in physiological regulation. An alternative total synthesis route for MS-444 was reported, starting from commercially available materials. Most reactions in the route are easily implemented and have the potential for large-scale application.
MS-444, as a myosin light chain kinase and HuR inhibitor, plays an important role in physiological reg-ulation. An alternative total synthesis of MS-444 is reported by a route incorporating Sonogashira cou-pling, addition/elimination of allenic sulfonium salts generated in situ and intramolecular Friedel-Crafts acylation. This approach starts from commercially available materials and involves a distinctive method of constructing furan ring. Most of reactions in the route are readily to implement and have the prospect of large-scale application. (c) 2021 Elsevier Ltd. All rights reserved.
