The 3-D conformational shape of N-naphthyl-cyclopenta[d]pyrimidines affects their potency as microtubule targeting agents and their antitumor activity

Summary: A series of methoxy naphthyl substituted cyclopenta [d]pyrimidine compounds were designed and synthesized to study the influence of 3-D conformation on microtubule depolymerizing and antiproliferative activities. The NOESY studies showed that the rotational flexibility and conformational dissimilarity between two compounds, 4 and 5, led to significant differences in biological activities. Compound 5 exhibited potent microtubule depolymerizing effects and strong antitumor activity, making it a potential preclinical candidate for further studies.

Some compounds showed potent COX-2 inhibition and in vivo anti-inflammatory activity, with certain derivatives displaying superior efficacy and safety profile compared to the reference drug celecoxib. Molecular docking studies indicated similar binding interactions of select compounds 10c, 10j, and 14e as selective COX-2 inhibitors, with a higher potential for accessing the selectivity side pocket.

Electronic transport through molecules containing pyrimidine units: First-principles calculations

The presence of pyrimidine rings enhanced the current through the junctions for both polarities of the applied voltage, showing clear current rectification due to the polar nature of the molecules. The effect of the dihedral angle between phenyl and pyrimidine rings on current rectification was also explored, with results explained in terms of charge localization in the system discovered through transmission eigenvalues analysis. These findings can aid in understanding the role of polar self-assembled monolayers in interface engineering.

Summary: A novel inhibitor, 2-benzylthiopyrimidine, was developed from 2-mercaptopyrimidine to inhibit the corrosion of carbon steel in CO2-containing environment, showing high inhibition efficiency in electrochemical measurements. Theoretical calculations indicated that the protonation of both precursors is rare, while the benzyl substituent in 2-benzylthiopyrimidine enhances its adsorption ability. Both inhibitors can adsorb on Fe surface in a nearly vertical orientation through the bonding of N and S atoms.
