Optogenetic activation of muscle contractionin vivo

Elahe Ganji et al.

Summary: This study aimed to evaluate the feasibility of non-invasive optogenetic stimulation of skeletal muscle in mice. By inducing specific gene expression in skeletal muscle and using transdermal blue light exposure for muscle contraction experiments, the study demonstrated the impact of optogenetic stimulation on sustained muscle contraction.

Bi-fated tendon-to-bone attachment cells are regulated by shared enhancers and KLF transcription factors

Shiri Kult et al.

Summary: The study found that murine tendon-to-bone attachment cells are bi-fated, expressing characteristics of both chondrocytes and tenocytes, regulated by KLFs transcription factors. These cells share accessible regions, exhibit enhancer signatures, and are regulated by KLFs in their differentiation process.

Tendinopathy

Neal L. Millar et al.

Summary: Tendinopathy is a complex pathology of the tendon characterized by pain, decline in function, and reduced exercise tolerance. Diagnosis is mainly clinical, but sometimes pain-provoking tests and imaging are necessary. Management includes exercise programmes, therapeutic modalities, and surgical interventions but their effectiveness is still unclear. Future research should focus on understanding key functional pathways in the clinical disease and improving rehabilitation protocols.

Loss of Smad4 in the scleraxis cell lineage results in postnatal joint contracture

Saundra Y. Schlesinger et al.

Summary: This study established a novel murine model of joint contracture by targeting Smad4 deletion in the tendon cell lineage. Smad4(ScxCre) mutants showed joint contracture shortly after birth, with severity increasing with age. Disruption of the BMP pathway in Alk3(ScxCre)/Alk6(null) mutants replicated many features of the joint contracture phenotype, suggesting a role for BMP signaling in tendon elongation and extracellular matrix accumulation.

A Neuroskeletal Atlas: Spatial Mapping and Contextualization of Axon Subtypes Innervating the Long Bones of C3H and B6 Mice

Madelyn R. Lorenz et al.

Summary: This study established comprehensive maps of sympathetic adrenergic and sensory peptidergic axons within and around the femur and tibia in mice, defining three distinct patterns of periosteal innervation. The results revealed a gradient of bone marrow axon density that increases along the length of the tibia and identified key regions of interest for neuroskeletal studies. This information serves as a guide for future studies and was compiled as a resource for the field as part of the NIH SPARC consortium.

In vitro temporal HIF-mediated deposition of osteochondrogenic matrix governed by hypoxia and osteogenic factors synergy

Isabel Calejo et al.

Summary: The study investigated the influence of hypoxia and osteogenic medium on cell phenotype modulation using an enthesis coculture model. It was found that hypoxia and osteoinduction promoted a temporal chondro/osteogenic pathway activation, and the presence of OM under hypoxia influenced the extracellular ROS/HIF1A interplay. This research revealed a link between biochemical factors and cell-cell crosstalk towards enthesis-like phenotypes.
