Evaluation of Histidine Reactivity and Byproduct Formation during Peptide Chlorination

Summary: The reactivity of histidine towards chlorine transformation products is lower compared to other chlorine-reactive amino acids, with chlorine preferentially targeting tyrosine and other more reactive amino acids. Over short time scales, the more reactive amino acids may contribute more to structural modifications to proteins. Over longer time scales, beta-cyanoalanine is identified as the dominant transformation product of chlorine reactions with peptide-bound histidine.
