Sustainability Challenges and Opportunities in Oligonucleotide Manufacturing

Summary: With a renewed and growing interest in therapeutic oligonucleotides across the pharmaceutical industry, pressure is increasing on drug developers to take more seriously the sustainability ramifications of this modality. Current state of the art in oligonucleotide production poses a waste and cost burden to manufacturers, leading to the identification of green chemistry processes as a critical unmet need. Collaborations between academia, research organizations, and the pharmaceutical industry are encouraged to address the challenges and improve sustainability in oligonucleotide production.

Stereochemistry Enhances Potency, Efficacy, and Durability of Malat1 Antisense Oligonucleotides In Vitro and In Vivo in Multiple Species

Summary: The study aimed to assess the impact of stereochemistry on the activity of antisense oligonucleotides delivered by intravitreal injection. Results showed that the stereopure oligonucleotide displayed superior potency, efficacy, and durability, with nearly 95% reduction of MALAT1 RNA in nonhuman primate eyes. This suggests that stereopure oligonucleotides have the potential to offer less frequent administration and low-dose regimens for patients with genetic eye diseases.

Summary: A static headspace capillary gas chromatography (HS-GC) method has been developed for the quantitative analysis of trace levels of acrylonitrile, featuring a short run time, wide calibration range, and a diluent with strong buffering and stabilizing capability. The method was validated for specificity, accuracy, repeatability/precision, calibration range, limits of quantitation and detection, solution stability, and robustness, showing quadratic performance over a wide range of concentrations. This method is deemed suitable as a platform test method for acrylonitrile determination in oligonucleotide manufacturing processes.
