Insight into epigenetics of human endometriosis organoids: DNA methylation analysis of HOX genes and their cofactors

Fereshteh Esfandiari et al.

Summary: This study evaluated and compared the methylation pattern of HOX clusters and HOX cofactors in normal, eutopic, and ectopic endometrial tissues, utilizing ectopic and eutopic endometriosis organoids as preclinical models. Most genes showed methylation alterations in ectopic/eutopic tissues and organoids, but a contrary methylation pattern was observed in 28 genes. The findings suggest that endometriosis organoids could serve as a novel preclinical model for studying the epigenetic mechanisms underlying endometriosis.

Disturbed progesterone signalling in an advanced preclinical model of endometriosis

Fereshteh Esfandiari et al.

Summary: Human endometriosis organoids were established from biopsies of women with and without endometriosis and showed aberrant expressions of endometriosis-related genes compared to control endometrium organoids. These endometriosis organoids exhibited a decreased response to progesterone, with significant downregulation of Progesterone receptor B (PRB) gene expression. Epigenetic mechanisms such as DNA hypermethylation at the PRB promoter were detected in eutopic but not ectopic organoids, implying other mechanisms like histone modifications and microRNAs may be responsible for PRB downregulation in ectopic organoids. Endometriosis organoids serve as valuable preclinical models for investigating the molecular mechanisms of endometriosis-associated progesterone resistance.

Endometriosis in Canada: It Is Time for Collaboration to Advance Patient-Oriented, Evidence-Based Policy, Care, and Research

Kate J. Wahl et al.

Summary: Endometriosis is a mysterious and complex disease that affects approximately 1 million people in Canada, causing chronic pain and infertility. Various factors can lead to diagnostic delays and limit access to care. An integrated approach involving collaboration among patients, clinicians, and researchers is urgently needed to improve awareness, best practices, and research coordination across the country.
