High-accuracy protein structure prediction in CASP14

Joana Pereira et al.

Summary: The application of state-of-the-art deep-learning approaches to protein modeling problem has expanded the high-accuracy category in CASP14, evaluating the performance of different groups and introducing new metrics. Despite the significant progress made by AlphaFold2, the second-best method in CASP14 outperformed the best method in CASP13, demonstrating the role of community-based benchmarking in the development of protein structure prediction field.

Assessing the utility of CASP14 models for molecular replacement

Claudia Millan et al.

Summary: This study introduces a new method (reLLG) for evaluating protein structure models that does not require diffraction data. Calibration against CASP14 targets showed that reLLG is a robust measure of model and group ranking. Additionally, refinements by CASP groups often lead to improved accuracy in models.

Topology evaluation of models for difficult targets in the 14th round of the critical assessment of protein structure prediction

Lisa N. Kinch et al.

Summary: This report summarizes the tertiary structure prediction assessment of difficult modeling targets in CASP14, with the top-performing AlphaFold2 group providing high quality models. Despite significant progress in protein structure prediction, challenges remain with flexible regions and obligate oligomeric assemblies. Performance-based PCA and heatmap clusters offer insight into target difficulties and successful structure prediction methodologies.

Critical assessment of methods of protein structure prediction (CASP)-Round XIV

Summary: CASP is a community experiment aimed at advancing methods for computing three-dimensional protein structure, including rigorous blind testing and evaluation by independent assessors. In the recent CASP14 experiment, deep-learning methods from one research group consistently delivered computed structures rivaling the corresponding experimental ones in accuracy. These results represent a solution to the classical protein-folding problem, at least for single proteins.
