Advances in Activity-Based Sensing Probes for Isoform-Selective Imaging of Enzymatic Activity

Sarah H. Gardner et al.

Summary: Activity-based sensing (ABS) has emerged as a powerful approach for monitoring small-molecule and enzyme activities within living systems, with a recent focus on increasing selectivity to monitor a single enzyme or isoform. The highest degree of selectivity is required for differentiating between isoforms, where the targets display significant structural similarities as a result of gene duplication or alternative splicing. This Minireview highlights key examples of small-molecule isoform-selective probes, focusing on the relevance of isoform differentiation, design strategies to achieve selectivity, and applications in basic biology or in the clinic.

Copper metabolism as a unique vulnerability in cancer

Vinit C. Shanbhag et al.

Summary: Over the past 25 years, significant progress has been made in identifying and characterizing proteins that regulate the uptake, intracellular trafficking and export of copper. While copper is essential for growth and development in humans and other mammals, it is also a limiting nutrient for cancer cell growth and proliferation. Oral copper chelators have been shown to have anti-neoplastic and anti-metastatic effects in animals and humans.

Development of NIR-II Photoacoustic Probes Tailored for Deep-Tissue Sensing of Nitric Oxide

Melissa Y. Lucero et al.

Summary: Photoacoustic imaging using near infrared I light has limitations due to interference from endogenous biomolecules, prompting the development of NIR-II imaging agents for improved sensitivity. The study introduced APNO-1080 as a NIR-II nitric oxide-responsive probe, demonstrating enhanced sensitivity and deep-tissue imaging capabilities in vitro and in vivo models of cancer.
