PARP inhibitor Olaparib overcomes Sorafenib resistance through reshaping the pluripotent transcriptome in hepatocellular carcinoma

Xiao-Dong Yang et al.

Summary: PARP1 plays important roles in stem cell pluripotency and HCC sorafenib treatment resistance. Overexpression of PARP1 is closely associated with poor clinical outcome in HCC patients. Olaparib suppresses DNA damage repair signaling and the global pluripotent transcriptional network, enhancing sorafenib in eliminating HCC residual tumors.

Chemotherapy-Enriched THBS2-Deficient Cancer Stem Cells Drive Hepatocarcinogenesis through Matrix Softness Induced Histone H3 Modifications

Kai-Yu Ng et al.

Summary: The study reveals the role of chemotherapy-enriched CD133+ liver cancer stem cells with THBS2 deficiency in promoting an aggressive hepatocellular carcinoma phenotype by remodeling the extracellular matrix. THBS2 deficiency is associated with low HCC survival and the findings have implications for cancer treatment.

Targeting tumor lineage plasticity in hepatocellular carcinoma using an anti-CLDN6 antibody-drug conjugate

Fan-En Kong et al.

Summary: Tumor lineage plasticity is increasingly recognized as a key mechanism of therapeutic resistance, where Claudin6 (CLDN6) was found to play a role in inducing a phenotypic shift in hepatocellular carcinoma (HCC) cells, making them more difficult to treat. Activation of the CLDN6/TJP2/YAP1 axis and Hippo signaling pathway may enhance tumor lineage plasticity and induce changes in cellular identity. Furthermore, a novel anti-CLDN6 monoclonal antibody conjugated with a cytotoxic agent showed potent antitumor efficacy in HCC treatment.

The interplay of UBE2T and Mule in regulating Wnt/β-catenin activation to promote hepatocellular carcinoma progression

Nicole Pui Yu Ho et al.

Summary: Recent evidence has shown the significant role of cancer stem cells (CSCs) in tumor relapse and therapeutic resistance in hepatocellular carcinoma (HCC) patients. A study identified UBE2T as a key gene in HCC, revealing its involvement in regulating liver CSCs, and suggesting it as a potential therapeutic target for HCC. The study also uncovered a novel UBE2T/Mule/beta-catenin signaling cascade that plays a critical role in the regulation of liver CSCs, highlighting its importance in HCC treatment.
