Genome-wide CRISPR/Cas9 deletion screen defines mitochondrial gene essentiality and identifies routes for tumour cell viability in hypoxia

Luke W. Thomas et al.

Summary: The study identified essential mitochondrial genes for tumor cell viability under different metabolic conditions through genome-wide CRISPR/Cas9 screening. Loss of oxidative phosphorylation genes improved tumor cell growth in hypoxia-glucose but reduced growth in normoxia. Furthermore, loss of genes involved in energy-consuming processes enhanced cell viability in both hypoxia-glucose and normoxia-galactose compared to normoxia-glucose.

Tarloxotinib Is a Hypoxia-Activated Pan-HER Kinase Inhibitor Active Against a Broad Range of HER-Family Oncogenes

Adriana Estrada-Bernal et al.

Summary: Tarloxotinib, a prodrug that utilizes tumor hypoxia to generate a potent pan-HER tyrosine kinase inhibitor, demonstrated selective delivery to tumor cells and effective inhibition of EGFR exon 20, ERBB2 mutations, and NRG1 fusions in cancer models. The concentrated presence of tarloxotinib-E in tumor tissues led to tumor regression and growth inhibition in mouse xenografts, with promising clinical responses observed in patients with relevant mutations.

Summary: This study demonstrates that ultra high single-dose radiation therapy (24 Gy SDRT) is more effective in improving local control of oligometastatic lesions compared to standard hypofractionated stereotactic body radiation therapy (3 x 9 Gy SBRT), leading to reduced rates of local recurrence and distant metastatic progression.

Mitochondrial stress induced by continuous stimulation under hypoxia rapidly drives T cell exhaustion

Nicole E. Scharping et al.

Summary: Continuous TCR signaling and hypoxia increase Blimp-1, suppressing mitochondrial reprogramming and generating reactive oxygen species, leading to T cell exhaustion. Mitigating T cell-intrinsic ROS and reducing tumor hypoxia can limit T cell exhaustion, synergizing with immunotherapy.

Mitochondrial Inhibitor Atovaquone Increases Tumor Oxygenation and Inhibits Hypoxic Gene Expression in Patients with Non-Small Cell Lung Cancer

Michael Skwarski et al.

Summary: This study presents the first clinical evidence that targeting tumor mitochondrial metabolism can reduce hypoxia and produce relevant antitumor effects at the mRNA level in NSCLC. Repurposing atovaquone for this purpose may improve treatment outcomes for NSCLC patients, as demonstrated by a significant reduction in hypoxic volume and downregulation of hypoxia-regulated genes in atovaquone-treated tumors. No atovaquone-related adverse events were reported, suggesting its potential as a safe and effective treatment option for NSCLC.

Superoxide produced by mitochondrial site IQ inactivates cardiac succinate dehydrogenase and induces hepatic steatosis in Sod2 knockout mice

Hoi-Shan Wong et al.

Summary: The study demonstrated the effectiveness of novel variants of S1QELs in vivo, showing that mitochondrial site I-Q plays a significant role in driving pathology in Sod2(-/-) mice, while site IIIQo does not significantly contribute to mitochondrial superoxide production in these mice. Treatment with S1QEL352 and S1QEL712 improved cardiac and hepatic pathologies in Sod2(-/-) mice, confirming the importance of superoxide production from mitochondrial site I-Q in hepatic steatosis.

Metabolic support of tumour-infiltrating regulatory T cells by lactic acid

McLane J. Watson et al.

Summary: T-reg cells exhibit broad heterogeneity in glucose metabolism within normal and transformed tissues, utilizing an alternative metabolic pathway to maintain their suppressive function and proliferation. Glucose uptake is correlated with poorer suppressive function and long-term instability, while high-glucose conditions impair T-reg cell function and stability. Additionally, T-reg cells upregulate pathways involved in the metabolism of lactic acid, with lactate uptake being dispensable for peripheral T-reg cells but required intratumorally for slowed tumor growth and increased response to immunotherapy.

Dan P. Zandberg et al.

Summary: The study analyzed the effect of anti-PD-1 therapy in patients with recurrent/metastatic squamous cell carcinoma of the head and neck (HNSCC), finding that anti-PD-1 resistance is associated with changes in tumor cell metabolism, increased tumor hypoxia, and decreased CD8+ T cell numbers. Lower tumor hypoxia was associated with increased efficacy of anti-PD-1 therapy in patients.

Summary: Hypoxia, a characteristic of the tumor microenvironment, has been shown to confer resistance to traditional chemotherapy but can synergize with PARP inhibitors under severe hypoxia. Moderate hypoxia, however, promotes resistance to PARP inhibitors. Selective elimination of hypoxic tumor cells enhances the efficacy of PARP inhibitor therapy without increasing normal tissue toxicity.

Linking EMT programmes to normal and neoplastic epithelial stem cells

Arthur W. Lambert et al.

Summary: Epithelial stem cells play critical roles in tissue generation, maintenance, and repair, while cancer stem cells are implicated in the growth and recurrence of many carcinomas. The activation of epithelial-mesenchymal transition (EMT) is linked to the formation of both normal and neoplastic stem cells, but the mechanistic basis of this connection remains unclear. Understanding the elusive links between EMT programs and the stem cell state will advance our knowledge of normal stem cell biology and cancer pathogenesis.

Endoplasmic reticulum stress signals in the tumour and its microenvironment
