From C4 to C7: Innovative Strategies for Site-Selective Functionalization of Indole C-H Bonds

Jian Wen et al.

Summary: The presence of hydrocarbons makes C-H functionalization an attractive alternative, with indole being an important heteroarene. Selective C-H functionalization at the benzene core of indoles is challenging, with efforts focusing on directing group development. Innovative strategies have been developed to selectively functionalize different positions of indoles, offering efficient methods for the synthesis of valuable substituted indoles.

Recent Advances in Metal-Catalyzed Functionalization of Indoles

Kelvin Urbina et al.

Summary: Indole is an important heterocycle in organic synthesis, natural products, and drug discovery, and recent research on the selective functionalization of indole has made significant advances, driven mainly by transition metal catalysis and metal-free methods. Future trends include transition-metal-catalyzed functionalization at different positions and transition-metal-free functionalization.

N-substituted aminobiphenyl palladacycles stabilized by dialkylterphenyl phosphanes: Preparation and applications in C-N cross-coupling reactions

Andrea Monti et al.

Summary: The study presented the synthesis and characterization of neutral and cationic N-methyl- and N-phenyl-2-aminobiphenyl methanesulfonate palladacycles stabilized with dialkylterphenyl phosphines. By selecting different phosphine ligands, different coordination modes and structures of complexes were obtained. The most efficient precatalyst, [Pd(N-methyl-2-aminobiphenyl)L4](OMs), was identified for facilitating the coupling of aryl chlorides with secondary and primary amines and indoles.

Summary: This review highlights the importance of remote regioselectivity in the metal-free organocatalytic functionalization of indoles in the carbocyclic ring positions up to the middle of 2020. Exciting developments have been achieved to access remote regioselectivities at the C4, C5, C6, and C7 positions of the indole core, overcoming the challenges posed by the dominant reactivity of the pyrrole core.

Recent developments in selective N-arylation of azoles

Pallabi Halder et al.

Summary: Transition-metal based carbon-heteroatom (C-X) bond formation has been a focus of attention for synthetic chemists in recent years, for the synthesis of important aryl/heteroaryl motifs. Efficient protocols such as Buchwald-Hartwig amination, Ullmann coupling, Chan-Lam coupling, and metal-free approaches have been utilized for selective arylation of one hetero-centre in substrates. Selective N-arylation on azole-containing frameworks, either on one N-centre or the exocyclic N-site of the azole ring, has also been explored.

Direct C2-arylation of N-acyl pyrroles with aryl halides under palladium catalysis

Weiqiang Chen et al.

Summary: A C2-arylation reaction of N-acyl pyrroles using Pd(PPh3)(4) as a catalyst in combination with Ag2CO3 under air has been developed for the first time. This method provides a straightforward approach to the synthesis of valuable arylated pyrroles with good substrate tolerance and moderate to good yields under standard conditions. Interestingly, substrates with a thiophene or furan ring were found to be more reactive than pyrrole in the present catalytic system.
