Small molecules targeting cyclooxygenase/prostanoid cascade in experimental brain ischemia: Do they translate?

Jianxiong Jiang et al.

Summary: Acute brain ischemia, a common cause of stroke, significantly impacts mortality and permanent disabilities in survivors. Current treatments such as intravenous thrombolysis and mechanical thrombectomy are limited by narrow treatment windows and potential risks. Innovative treatment strategies are needed to improve outcomes for patients with acute brain ischemia.

ACSL4 exacerbates ischemic stroke by promoting ferroptosis-induced brain injury and neuroinflammation

Yu Cui et al.

Summary: ACSL4 is an important regulator of neuronal death and neuroinflammation in ischemic stroke, with its suppression protecting mice against brain ischemia and promoting neuronal death via enhancing lipid peroxidation. Additionally, knockdown of ACSL4 can inhibit proinflammatory cytokine production in microglia.

Cottonseed Oil Alleviates Ischemic Stroke-Induced Oxidative Stress Injury Via Activating the Nrf2 Signaling Pathway

Min Liu et al.

Summary: The study demonstrated that cottonseed oil has a positive effect on oxidative stress injury induced by ischemic stroke. Treatment with CSO significantly improved neurological deficit, reduced infarction volume, increased antioxidant activity, and activated the Nrf2 signaling pathway in male rats. Additionally, in neuronal cells, CSO treatment increased cell viability, decreased cell apoptosis, and induced Nrf2 nuclear translocation after oxygen and glucose deprivation injury.

Palmatine Protects against Cerebral Ischemia/Reperfusion Injury by Activation of the AMPK/Nrf2 Pathway

Chaoliang Tang et al.

Summary: Palmatine (PAL), a natural isoquinoline alkaloid, exhibits extensive biological and pharmaceutical activities, including antioxidative stress, anti-inflammatory, antitumor effects. PAL treatment shows protective effects against ischemic stroke and cerebral ischemia/reperfusion injury by attenuating oxidative stress, inflammatory response, and neuronal apoptosis, possibly through the activation of the AMP-activated protein kinase (AMPK)/nuclear factor E2-related factor 2 (Nrf2) pathway.

Sinomenine activation of Nrf2 signaling prevents inflammation and cerebral injury in a mouse model of ischemic stroke

Fangfang Bi et al.

Summary: Sinomenine (SINO), derived from Sinomenium acutum, is clinically used for treating rheumatoid arthritis and neuralgia. It exerts anti-inflammatory and cerebral protective effects possibly through regulating the Nrf2 signaling pathway. The study showed that SINO upregulated Nrf2 protein expression, reduced cerebral damage and inflammation in a middle cerebral artery occlusion model, highlighting its potential in treating cerebral inflammatory disorders.

Pelargonidin ameliorates MCAO-induced cerebral ischemia/reperfusion injury in rats by the action on the Nrf2/HO-1 pathway

Kong Fu et al.

Summary: This study demonstrated that pelargonidin could effectively reduce cerebral ischemia volume, improve neurological function, and enhance memory and learning ability in MCAO rats. The mechanism of action may be related to the regulation of the Nrf2/HO-1 signaling pathway.
