Interaction between GALNT12 and C1GALT1 Associates with Galactose-Deficient IgAl and IgA Nephropathy

Summary: The study identified two novel loci, C1GALT1 and GALNT12, associated with serum levels of galactose-deficient IgA1 in patients with IgA nephropathy. The genetic interaction between GALNT12 and C1GALT1 was found to be significant, suggesting a key role of genetically conferred dysregulation of galactose-deficient IgA1 in the development of IgA nephropathy. Additionally, GALNT12 mRNA expression was significantly lower in patients with IgA nephropathy compared to healthy controls.

Global functions of O-glycosylation: promises and challenges in O-glycobiology

Summary: O-glycosylation, particularly O-GalNAc glycans, plays crucial roles in tissue development, homeostasis, and host-microbe interactions. Technological advancements, such as mass spectrometry and genetic engineering, have allowed for a better understanding of O-glycosylation profiles and functions, with high-throughput O-glycoproteomics providing comprehensive insights. However, there is still much to explore, especially in understanding the context-dependent functions of O-glycans in different cellular processes and disease states.

It was found that the luminal stem region and catalytic domain of ppGalNAc-T18 are essential for ER localization, while the lectin domain and N-glycosylation are not required. The findings provide insights into the subcellular localization mechanism of ppGalNAc-T18 and its role in non-catalytic O-GalNAc glycosylation activity.
