Summary: In this study, a system for enhancing the extracellular secretion of the enzyme TM1 in Escherichia coli was developed. Utilizing the signal recognition particle (SRP)-dependent translocation pathway, the co-translational extracellular secretion of TM1 (D-TM1) resulted in significantly higher specific activity compared to the PelB signal peptide (P-TM1) overexpression. This system shows potential for enhancing E. coli's capacity as a microbial cell factory for extracellular secretion of a variety of important proteins.
