Excited state structure of isolated 4-cyanoindole from a combined Franck-Condon and rotational constants analysis

Summary: The changes in the structure of 4-cyanoindole upon electronic excitation were determined through a combined Franck-Condon method and rotational constants fitting. These geometry changes were fitted to simultaneously reproduce the intensities of 72 lines in the fluorescence emission spectra, obtained through excitation of six different vibronic bands in the first excited singlet state, along with the three rotational constants obtained from rotationally resolved electronic spectra.

A Scalable Synthesis of the Blue Fluorescent Amino Acid 4-Cyanotryptophan and the Fmoc Derivative: Utility Demonstrated with the Influenza M2 Peptide Tetramer

Summary: A scalable synthesis of a Fmoc-protected blue fluorescent amino acid, L-4-cyanotryptophan (W-4CN), was reported in this study. The study leveraged the red-shifted emission of water-exposed W-4CN residues to investigate the solvation state of tryptophan residues within the influenza M2 proton channel. The correlation between the conformation of the channel and the fluorescence spectrum of a mutated W-4CN residue suggested that the channel's conformational state does not affect the hydration status of the Trp residues.

Tuning the electronic transition energy of indole via substitution: application to identify tryptophan-based chromophores that absorb and emit visible light

Summary: The absorption spectra of a library of 4-substituted indoles were examined, revealing a linear correlation between the frequency of the absorption maximum and the global electrophilicity index of the substituent. This allowed the identification of two promising candidates, 4-formyltryptophan and 4-nitrotryptophan, both of which can be excited by visible light. While 4-formyltryptophan emits cyan fluorescence with a reasonably large quantum yield, 4-nitrotryptophan is essentially non-fluorescent, suggesting potential use as a fluorescence reporter or quencher.

Summary: Recent progress on STED fluorophores for super-resolution imaging is outlined, including inorganic fluorophores, fluorescent proteins, organic luminescent materials, aggregation-induced emission (AIE) luminogens, and fluorescent nanoparticles. Characteristics of these aforementioned STED fluorophores are also included and compared to provide a deep understanding of the relationship between the properties in luminescent materials and their performance in STED imaging.
