Allopregnanolone and Pregnanolone Are Reduced in the Hippocampus of Epileptic Rats, but Only Allopregnanolone Correlates with Seizure Frequency

Chiara Lucchi et al.

Summary: The study found decreased levels of allopregnanolone and pregnanolone in the hippocampus of epileptic rats, which were positively correlated with seizure frequency, while no changes were found in the neocortex.

Intranasal Allopregnanolone Confers Rapid Seizure Protection: Evidence for Direct Nose-to-Brain Delivery

Dorota Zolkowska et al.

Summary: Allopregnanolone administered intranasally rapidly elevates seizure threshold with minimal adverse effects, showing direct nose-to-brain delivery and preferential distribution in brain regions relevant to seizures compared to benzodiazepines. Intranasal delivery of allopregnanolone provides a rapid and effective treatment option for seizures, overcoming potential refractoriness to benzodiazepines.

Looking for Novelty in an Old Receptor: Recent Advances Toward Our Understanding of GABAARs and Their Implications in Receptor Pharmacology

Summary: GABA(A) receptors play a crucial role in neurological and neuropsychiatric disorders, and current pharmacological interventions targeting these receptors have limitations in terms of selectivity and side effects. The development of more selective drugs and exploration of next-generation drugs targeting accessory proteins associated with the receptors are necessary in order to improve treatment outcomes.

Astroglial glutamate transporter 1 and glutamine synthetase of the nucleus accumbens are involved in the antidepressant-like effects of allopregnanolone in learned helplessness rats

Masahito Nangaku et al.

Summary: Clinical studies have shown that allopregnanolone has antidepressant-like effects on patients with depression. This study aimed to investigate the involvement of astroglial GLT-1 and GS in the antidepressant-like effects of ALLO in LH rats. The results indicated a connection between the antidepressant-like effects of ALLO and the astroglial GLT-1/GS system in the NAc region of LH rats.

Neurosteroids and Neurotrophic Factors: What Is Their Promise as Biomarkers for Major Depression and PTSD?

Felipe Borges Almeida et al.

Summary: This paper explores the potential of neurosteroids and neurotrophic proteins as biomarkers for MDD and PTSD. Findings show that allopregnanolone levels are diminished in MDD and PTSD patients, while BDNF is reduced in these patients and depressed-like animals with lower neurosteroid levels. The proposed causal link between allopregnanolone levels and BDNF expression could serve as a biomarker axis for the diagnosis of both MDD and PTSD.

(3α,5α)3-hydroxypregnan-20-one (3α,5α-THP) regulation of hypothalamic and extrahypothalamic corticotropin releasing factor (CRF): Sexual dimorphism and brain region specificity in Sprague Dawley rats

Giorgia Boero et al.

Summary: CRF is the main activator of the HPA axis in response to stress, with CRF neurons mainly located in the hypothalamus. 3 alpha,5 alpha-THP attenuates the stress response by reducing hypothalamic CRF mRNA expression and serum corticosterone levels. Sex and region-specific regulation of CRF signals in response to 3 alpha,5 alpha-THP administration may be essential for developing therapies for stress-related disorders and addiction.

Neurosteroid allopregnanolone (3α,5α-THP) inhibits inflammatory signals induced by activated MyD88-dependent toll-like receptors

Irina Balan et al.

Summary: The research shows that 3 alpha, 5 alpha-THP inhibits the activation of TLR4, TLR2, and TLR7 signals, but not TLR3 signal, by blocking the TLR-MyD88 interaction. This inhibition leads to decreased production of pro-inflammatory mediators and supports the potential therapeutic use of 3 alpha, 5 alpha-THP for inflammatory diseases.
