Somatic mosaicism in the MAPK pathway in sporadic brain arteriovenous malformation and association with phenotype

Sen Gao et al.

Summary: This study confirmed the high prevalence of somatic KRAS mutations in sporadic BAVM lesions and identified several candidate somatic variants in other MAPK pathway genes. These somatic variants may contribute to understanding the etiology of sporadic BAVM and the clinical characteristics of patients with this condition.

Summary: The study demonstrates that multi-slice SE-EPI aortic MRE using an FB protocol or a single-BH protocol is reproducible and repeatable with similar stiffness measurements compared to FB GRE. The FB SE-EPI measurements are highly reproducible and consistent, even across different scanners. Additionally, single-BH SE-EPI scans show a low coefficient of variation and are comparable to FB SE-EPI scans.

Selective Endothelial Hyperactivation of Oncogenic KRAS Induces Brain Arteriovenous Malformations in Mice

Eun S. Park et al.

Summary: Brain arteriovenous malformations (bAVMs) are a major cause of hemorrhagic stroke and neurological deficits in children and young adults, with no pharmacological intervention available. KRAS mutations, commonly seen in cancer, have been found in human sporadic bAVMs, leading to a new direction in research. Using AAV-BR1, the study demonstrated that endothelial KRAS(G12V) mutation induces bAVMs in mice, and MEK/ERK inhibition can suppress bAVM growth.

Proof-of-concept single-arm trial of bevacizumab therapy for brain arteriovenous malformation

Rachel Muster et al.

Summary: Brain arteriovenous malformations (bAVMs) are rare but require essential diagnosis and management due to the risk of secondary intracranial hemorrhage (ICH). Invasive therapies are currently the main interventions for reducing ICH risk. Bevacizumab has shown potential in reducing AVM size, as seen in this pilot study with two patients. More clinical trials are needed to assess its efficacy and potential adverse effects.

Summary: This study compared neurological outcomes in patients with grade III AVMs who underwent preoperative embolization or not, and found significant differences in outcomes. Patients who underwent embolization had fewer poor outcomes, indicating the potential benefits of preoperative embolization for grade III AVMs.
