Reticulon-3 Promotes Endosome Maturation at ER Membrane Contact Sites

Haoxi Wu et al.

Summary: The study reveals that the reticulon protein Rtn3L enriches at ER-endosome MCSs during endosome maturation, recruited by endosomal protein Rab9a. Depletion or deletion of Rtn3L leads to endosome maturation and cargo sorting defects, similar to RAB9A depletion.

An Arf/Rab cascade controls the growth and invasiveness of glioblastoma

Gopinath Kulasekaran et al.

Summary: The study demonstrated that loss of function of the endosomal GTPase Rab35 in human brain tumor initiating cells increases glioblastoma growth and decreases animal survival. Mechanistically, the GTPase Arf5 enhances its GEF activity toward Rab35, resulting in increased growth and invasiveness of BTIC-initiated brain tumors.

Membrane Protein Quality Control Mechanisms in the Endo-Lysosome System

Richa Sardana et al.

Summary: Protein quality control machineries are essential for identifying and removing mistargeted, misfolded, and aberrant proteins, but the complexity of the proteome makes this task challenging. Genetic and proteomic approaches are beginning to reveal how these machineries recognize targets and regulate their activity. This review provides an overview of E3 ubiquitin ligases and associated machinery involved in protein quality control, aiming to guide future research in this area.

Mechanisms of nonvesicular lipid transport

Karin M. Reinisch et al.

Summary: Lipids can be transported between cellular membranes via nonvesicular mechanisms, supporting biogenesis of rapidly expanding membranes or allowing targeted modulation of specific lipid levels. Nonvesicular transport likely occurs at membrane contact sites and involves lipid transport proteins, but many questions remain unanswered regarding the mechanisms involved.

Structural basis for VPS34 kinase activation by Rab1 and Rab5 on membranes

Shirley Tremel et al.

Summary: The lipid phosphatidylinositol-3-phosphate (PI3P) acts as a regulator of endocytosis and autophagy, generated by two complexes containing the lipid kinase VPS34. Rab5 and Rab1a GTPases recruit and activate specific VPS34 complexes on membranes through unique binding interfaces, guiding them to distinct cellular locations. Insights into the recruitment and activation mechanisms of VPS34 complexes by specific Rab GTPases are provided through biochemical and structural analyses.

Summary: Rab5 and Rab7a play crucial roles in regulating endosomal progression and maturation. The switch from Rab5 to Rab7a induces the formation of new fully functional early endosomes, which in turn regulates the direction of transport and the homeostasis of early endosomes.

Cryo-EM structure of metazoan TRAPPIII, the multi-subunit complex that activates the GTPase Rab1

Antonio Galindo et al.

Summary: TRAPP complexes, which are nucleotide exchange factors, play vital roles in membrane traffic and autophagy. TRAPPII activates Rab11, while TRAPPIII activates Rab1, with shared core subunits affecting nucleotide exchange and specific large subunits essential for in vivo activity. Cryo-EM structures reveal the assembly of TRAPPIII complex, showing how TRAPPIII-specific subunits hold the catalytic core and large arms determine specificity. The lower resolution structure of TRAPPII suggests evolution from a single ur-TRAPP.

The Rab7 effector WDR91 promotes autophagy-lysosome degradation in neurons by regulating lysosome fusion

Ruxiao Xing et al.

Summary: WDR91, a Rab7 effector crucial for early-to-late endosome conversion, plays a vital role in maintaining lysosome function and homeostasis. Lack of WDR91 in the central nervous system results in neuronal loss, behavioral defects, and abnormal lysosomes, leading to accumulation of autophagic cargoes in mouse neurons. WDR91 competes with VPS41 for Rab7 binding, facilitating controlled lysosome fusion and ensuring normal function and survival of neurons.
