Prognostic significance of esterase gene expression in multiple myeloma

Summary: This study evaluated the biological significance of esterase expression in multiple myeloma (MM) by analyzing gene expression profiling and genomic variants in patients with newly diagnosed MM (NDMM) or relapsed/refractory MM (RRMM). High or low expression of specific esterases in MM, such as UCHL5, SIAE, ESD, PAFAH1B3, PNPLA4, and PON1, were significantly altered as patients progressed from NDMM to RRMM. Certain esterases, like OVCA2, PAFAH1B3, SIAE, USP4, and PCED1B, were identified as poor prognostic markers, suggesting their potential role in myeloma biology.

Synthetic O-acetylated sialosides facilitate functional receptor identification for human respiratory viruses

Summary: The study found specificities of human and animal viruses in binding to O-acetylated sialic acid. A chemoenzymatic methodology was developed to provide almost any sialate-acetylation pattern, and a microarray of O-acetylated sialoglycans was used to determine receptor specificity. Host-specific patterns of receptor recognition were observed, with human coronaviruses found to uniquely bind to a specific glycotope as part of a ganglioside.

Probing the binding specificities of human Siglecs by cell-based glycan arrays

Summary: Siglecs are a family of sialic acid-binding receptors expressed by immune cells, which exhibit distinct binding preferences for sialylated GalNAc-type O-glycans presented on different protein sequences. Sulfotransferases can influence Siglec-sialoglycan interactions and impact the binding preferences for O-glycan patterns, highlighting a potential link between Siglec-3 binding epitope and late-onset Alzheimer's disease. The study of human sialome in cell-based display provides a versatile platform for dissecting the genetic and biosynthetic basis of the Siglec glycan interactome and other sialic acid-binding proteins.

Display of the human mucinome with defined O-glycans by gene engineered cells
