Phase-selective staining of model and cell membranes, lipid droplets and lipoproteins with fluorescent solvatochromic pyrene probes

Summary: The push-pull solvatochromic pyrene derivatives PA and PK have been successfully applied as polarity-sensitive probes in the study of model membrane vesicles, cells and purified human serum lipoproteins. These probes provide information about lipid order in biomembranes, with the advantage of being excitable by a standard 405 nm laser light. The results from the study showed the capability of PA and PK to report differently on liquid-disordered, liquid-ordered and gel phase bilayers.

Recent progresses in fluorescent probes for detection of polarity

Fluorescent probes are commonly used to detect polarity changes, offering simplicity, selectivity, sensitivity, real-time monitoring, non-invasion, and high temporal-spatial resolution. Design strategies, response mechanisms, and imaging applications of organic fluorescent probes have played a significant role in the detection of polarity changes.

Targeted Solvatochromic Fluorescent Probes for Imaging Lipid Order in Organelles under Oxidative and Mechanical Stress

Summary: A series of solvatochromic probes have been designed for specific targeting of different organelles in live cells, allowing the detection of variations in lipid order and quantitative description of lipid order in each organelle. These probes revealed the characteristics of organelles under stress conditions, with mitochondria and lysosomes particularly sensitive to stress. The new probes provide a powerful toolbox for monitoring cellular response to physical and chemical stimuli at the level of individual organelle membranes.
