Common genetic variants and modifiable risk factors underpin hypertrophic cardiomyopathy susceptibility and expressivity

Andrew R. Harper et al.

Summary: The study identified 12 genome-wide significant susceptibility loci for HCM, showing a strong polygenic influence, especially for sarcomere-negative HCM. A genetic risk score influenced the odds of HCM and phenotypic severity in carriers of sarcomere variants. Mendelian randomization identified diastolic blood pressure as a key modifiable risk factor for sarcomere-negative HCM.

Shared genetic pathways contribute to risk of hypertrophic and dilated cardiomyopathies with opposite directions of effect

Rafik Tadros et al.

Summary: The study found a strong genetic correlation between hypertrophic and dilated cardiomyopathies and left ventricular traits, with opposing genetic effects in the two cardiomyopathies. The research also supported a causal association between increased left ventricular contractility and hypertrophic cardiomyopathy risk.

Summary: This study analyzed tissue from patients with obstructive hypertrophic cardiomyopathy and heart failure to develop individualized protein-protein interaction networks using RNA-Seq technology. The findings showed that this approach can distinguish between different types of cardiomyopathy and provide key insights into patient-level pathobiology and clinically relevant characteristics of complex diseases.

Hypertrophic Cardiomyopathy in Children: Pathophysiology, Diagnosis, and Treatment of Non-sarcomeric Causes

Emanuele Monda et al.

Summary: Hypertrophic cardiomyopathy is characterized by left ventricular hypertrophy in children, with a diverse range of causes including sarcomeric gene mutations and non-sarcomeric causes. The age of onset and underlying etiology significantly impact the prognosis of children with HCM, and targeted approaches for non-sarcomeric etiologies have emerged in recent years. Etiological diagnosis is crucial in designing specific management and therapy for these rare disorders in children.

A method for cryopreservation and single nucleus RNA-sequencing of normal adult human interventricular septum heart tissue reveals cellular diversity and function

Summary: Single cell sequencing of human heart tissue is challenging due to technical difficulties and lack of standardized methods for cryopreservation. This study developed a reliable method for tissue mincing and cryopreservation, enabling high quality single nuclei RNA recovery. The method was successfully applied to normal human heart tissue, showing unexpected cellular diversity in the interventricular septum.

Summary: This study identified plasma protein biomarkers of HCM and revealed upregulated molecular pathways, such as the Ras-MAPK and TGF-beta pathways, in HCM patients. This investigation represents the largest-scale proteomics profiling in HCM, highlighting both novel and known pathways differentially regulated in the disease.
