Heterozygous disruption of ALAS1 in mice causes an accelerated age-dependent reduction in free heme, but not total heme, in skeletal muscle and liver

Summary: This study found an age-dependent reduction in free heme levels in the skeletal muscle and liver of ALAS1 heterozygous mice, while there was only a slight decrease in the liver of WT mice. Total heme levels did not show significant differences between young and aged mice. ALAS1 mRNA levels exhibited an age-dependent reduction similar to the free heme levels.

Restoring metabolism of myeloid cells reverses cognitive decline in ageing

Summary: Ageing is characterized by chronic pro-inflammatory responses that affect various aspects of health, including the brain. The study shows that in ageing mice, myeloid cell bioenergetics are suppressed in response to increased signalling by the lipid messenger prostaglandin E-2 (PGE(2)), leading to maladaptive pro-inflammatory responses. By inhibiting myeloid EP2 signalling, it is possible to reverse cognitive ageing and restore youthful immune functions.

This highlights the important role of Treg-derived IL-10 in regulating adipose tissue homeostasis.
