ROR1 targeting with the antibody-drug conjugate VLS-101 is effective in Richter syndrome patient-derived xenograft mouse models

Tiziana Vaisitti et al.

Summary: The Richter syndrome (RS) is a transformation of chronic lymphocytic leukemia (CLL) into an aggressive lymphoma, with limited treatment options and high fatality rates. Targeting the receptor tyrosine kinase-like orphan receptor 1 (ROR1) with the antibody-drug conjugate (ADC) VLS-101 has shown promising results in RS patient-derived xenografts, indicating the therapeutic potential of this approach for hematological cancers. A phase 1 clinical trial of VLS-101 (NCT03833180) is currently ongoing in patients with RS and other hematological malignancies.

Phase 1 study of single-agent WNT974, a first-in-class Porcupine inhibitor, in patients with advanced solid tumours

Jordi Rodon et al.

Summary: The Phase 1 study evaluated the safety and efficacy of Porcupine inhibitor WNT974 in patients with advanced solid tumours. The recommended dose for expansion was 10 mg once-daily, with dysgeusia being the most common adverse event. The study suggested that WNT974 may influence immune cell recruitment to tumours and enhance checkpoint inhibitor activity.

Targeting of Deregulated Wnt/β-Catenin Signaling by PRI-724 and LGK974 Inhibitors in Germ Cell Tumor Cell Lines

Silvia Schmidtova et al.

Summary: The majority of patients with testicular germ cell tumors can be cured with cisplatin-based chemotherapy, but for a subset with cisplatin-refractory disease, the prognosis is poor and treatment options are limited. Inhibitors of Wnt/beta-catenin signaling show promise in managing cisplatin-resistant GCTs, with PRI-724 demonstrating effectiveness in resistant cell lines. Further evaluation of Wnt/beta-catenin pathway inhibition in GCTs is warranted.
