Notch signaling in cancer: Complexity and challenges on the path to clinical translation

Antonio Ferreira et al.

Summary: Notch receptors participate in a conserved signaling pathway that has diverse effects on normal cells and is highly dependent on the cellular context and signaling dosage. Notch plays different roles in various types of cancers, acting as either an oncogene or a tumor suppressor and affecting tumor cells and stromal elements in multiple ways. This review provides an overview of the roles of Notch in cancer and discusses the challenges and potential approaches for clinical translation of Notch targeting agents.

Tetraspanins: useful multifunction proteins for the possible design and development of small-molecule therapeutic tools

Jean-Michel H. Robert et al.

Summary: Tetraspanins are a well-conserved superfamily of small membrane proteins in humans with important roles in cell biology, including cellular development, proliferation, activation, motility, and membrane fusions. Due to their diverse physiological functions, tetraspanins are considered regulators of cellular signaling and may play important roles in pathological processes.

Cryo-EM structure of the B cell co-receptor CD19 bound to the tetraspanin CD81

Katherine J. Susa et al.

Summary: The structure of the CD19-CD81 complex bound to a therapeutic antigen-binding fragment was determined by cryo-electron microscopy, revealing the contact interface between the ectodomains and the conformational changes of CD81 upon binding to CD19. This provides insights into the assembly of the CD19-CD81 complex and offers a basis for designing therapies for B cell dysfunction.

Dynamic Plasma Membrane Organization: A Complex Symphony

Sjoerd van Deventer et al.

Summary: The organization of membrane proteins is influenced by both intrinsic and extrinsic factors, leading to a dynamic exchange between protein monomers, clusters, and structures. Tetraspanin nanodomains are key organizers that are often overlooked, playing a crucial role in plasma membrane biology. Understanding how different key organizers collaborate is essential for a comprehensive grasp of membrane protein organization.

Tspan5 promotes epithelial-mesenchymal transition and tumour metastasis of hepatocellular carcinoma by activating Notch signalling

Qian Xie et al.

Summary: High expression of Tspan5 is significantly associated with clinicopathological features and poor survival in HCC patients, promoting cell migration and tumour metastasis. Mechanistically, Tspan5 enhances ADAM10 enzymatic activity, activates Notch signaling, and induces epithelial-mesenchymal transition in tumour cells. These findings highlight the pivotal role of Tspan5 in regulating Notch signaling, EMT, and tumour metastasis in HCC.
