Structural analysis of RIG-I-like receptors reveals ancient rules of engagement between diverse RNA helicases and TRIM ubiquitin ligases

Kazuki Kato et al.

Summary: The study uncovers evolutionarily conserved rules of engagement between RNA helicases and TRIM E3 ligases, showing their functional coordination in vertebrate innate immunity. These interactions are driven by RLR multimers, restricting the actions of TRIM proteins and consequent immune activation. Mass spectrometry and phylogeny-guided biochemical analyses suggest deep evolutionary connections between TRIM proteins and RNA helicases, linking ubiquitin and RNA biology throughout animal evolution.

RNF39 mediates K48-linked ubiquitination of DDX3X and inhibits RLR-dependent antiviral immunity

Wenwen Wang et al.

Summary: RLRs serve as major cytosolic RNA sensors in antiviral innate immunity, and their activation must be tightly controlled to prevent detrimental effects. This study reveals that the E3 ubiquitin ligase RNF39 inhibits RLR pathways by promoting K48-linked ubiquitination and proteasomal degradation of the key adapter DDX3X. This newly identified mechanism highlights RNF39 as a potential therapeutic target for diseases related to aberrant RLR activation.
