A Comparative Study of the Hybridization of Phosphoryl Guanidine Oligonucleotides with DNA and RNA

Summary: This study investigated the structure and thermal stability of complexes of RNA and DNA with phosphoryl guanidine oligonucleotides (PGO) bearing modified phosphate residues. It was found that substituting the internucleoside phosphate residue of an oligodeoxyribonucleotide with an electroneutral guanidine residue did not significantly alter the conformation of the PGO/RNA duplex. The introduction of modifications led to a decrease in thermal stability of PGO/RNA complexes.

Problems of the Synthesis of Oligonucleotide Derivatives in the Realization of the Anchimeric Effect

Summary: The presence of a free OH group at the 3'-terminal residue of the substituted ethylene glycol fragment affects the instability of the structure of phosphoryl guanidine oligodeoxyribonucleotide derivative. The by-products of the OH group's anchimeric effect include transesterification products of the PG unit. Mass spectrometry analysis indicates accumulation of derivatives without N,N,N',N'-substituted guanidine residue or the entire 3'-terminal PG-containing nonnucleotide unit under alkaline conditions.
