The neutrophil antimicrobial peptide cathelicidin promotes Th17 differentiation

Summary: The host defence peptide cathelicidin is a potent Th17 potentiator, enhancing aryl hydrocarbon receptor and ROR gamma t expression in a TGF-beta 1-dependent manner, directing T cells towards Th17 phenotype while protecting them from apoptosis. Neutrophils secrete cathelicidin, which is required for maximum Th17 differentiation and plays a role in early neutrophilia directing subsequent adaptive immune responses.

N beta neutrophils overexpress the alpha 4 beta 1 integrin compared to N alpha. Inhibiting alpha 4 beta 1 integrin can increase the N beta/N alpha ratio and enhance CD8 T-cell responses to HIV VACV-delivered antigens.

Summary: Women with SLE have higher proportions of activated LDG cells in peripheral and intervillous blood, as well as increased levels of CXCL8 and CXCL1 in the placenta compared to healthy pregnancies. IFN alpha may play a role in SLE pregnancy with detectable levels in intervillous blood and upregulating CXCL8 gene expression in decidual stromal cells. Chorioamnionitis was more common in SLE placentas, indicating a potential contribution to placental inflammation and dysfunction.
