Novel inhibitors with sulfamethazine backbone: synthesis and biological study of multi-target cholinesterases and α-glucosidase inhibitors

Cuneyt Turkes et al.

Summary: A series of novel ureido-substituted derivatives with sulfamethazine backbone were investigated for the inhibition of AChE, BChE, and alpha-GLY. Among these derivatives, compound 2c showed the most potent inhibition profile towards AChE and BChE. The study suggests the potential use of inhibitors with sulfamethazine backbone as innovative anticholinergic and antidiabetic agents.

From combinations to multitarget-directed ligands: A continuum in Alzheimer's disease polypharmacology

Claudia Albertini et al.

Summary: Despite the failures in drug discovery for complex neurodegenerative diseases like AD, the rise of awareness regarding polypharmacological approaches, using pharmaceutical agents acting on multiple targets, presents a promising avenue for restoring the diseased network and providing disease-modifying effects. Understanding the advantages and drawbacks of different modalities in drug discovery is crucial for developing successful AD multitarget drug-discovery endeavors. Researchers aiming to enter and progress in this field are encouraged to explore the full potential of AD polypharmacology.

Synthesis, Characterization, and Inhibition Study of Novel Substituted Phenylureido Sulfaguanidine Derivatives as α-Glycosidase and Cholinesterase Inhibitors

Suleyman Akocak et al.

Summary: A series of novel ureido-substituted sulfaguanidine derivatives were synthesized and their inhibitory effects on alpha-glycosidase, acetylcholinesterase, and butyrylcholinesterase enzymes associated with diabetes mellitus and Alzheimer's disease were investigated. The compounds showed higher antidiabetic effects compared to their anti-Alzheimer's effects, with stronger inhibition on the diabetic enzyme alpha-GLY than on esterase enzymes. Inhibition of metabolic enzymes is important for the treatment of DM and AD.
