Ubiquitin signalling in neurodegeneration: mechanisms and therapeutic opportunities

Summary: Neurodegenerative diseases are characterized by progressive nervous system damage, such as the selective loss of vulnerable neurons, leading to motor symptoms and cognitive decline. The primary risk factor for these diseases is aging, which results in reduced proteasomal degradation and autophagy, leading to the accumulation of neurotoxic protein aggregates. Ubiquitin plays a critical role in both degradation pathways and non-degradative signaling, implicating it as a potential target for new drugs to treat neurodegenerative diseases.

Knockdown of TRAP1 promotes cisplatin-induced apoptosis by promoting the ROS-dependent mitochondrial dysfunction in lung cancer cells

Summary: This study found that the expression of TRAP1 was significantly increased in cisplatin-induced lung cancer cells, knocking down TRAP1 induced apoptosis and G2/M cell cycle arrest in A549/DDP and H1299/DDP cells. It was suggested that TRAP1 suppressed cisplatin-induced apoptosis by promoting ROS-dependent mitochondrial dysfunction.

Insights Into the Mechanism of Tyrosine Nitration in Preventing β-Amyloid Aggregation in Alzheimer's Disease

Summary: Nitration of tyrosine at the tenth residue in amyloid-beta has been found to reduce its aggregation by inducing deprotonation of the phenolic hydroxyl group, resulting in a negatively charged Tyr10 at physiological pH. This negatively charged Tyr10 can interact with specific amino acids in Aβ and block the kink around Tyr10, preventing Aβ fibrillization and aggregation. These findings shed light on the importance of maintaining nitric oxide balance in vivo for preventing Alzheimer's disease.
