Expression, purification and characterization of the RhoA-binding domain of human SHIP2 in E.coli

Heng Zhou et al.

Summary: This study successfully designed, expressed, and purified two truncated proteins, SHIP2(118-298) and SHIP2(176-298), which were found to exist as homo-dimers and capable of binding with GTP-bound RhoA. The shorter truncation, SHIP2(176-298), was identified to contain the essential residues and may serve as a sufficient domain for further studies on the structure and RhoA binding of SHIP2. This work provides valuable information for understanding the interaction between SHIP2 and RhoA.
