Per- and Polyfluoroalkyl Substance Toxicity and Human Health Review: Current State of Knowledge and Strategies for Informing Future Research

Summary: Reports on the impacts of per- and polyfluoroalkyl substances (PFAS) on the environment and human health have increased substantially in peer-reviewed literature. Epidemiological studies have shown associations between exposure to specific PFAS and various health effects, and further research is needed to understand differences in responses between sexes, species, and life stages. More contemporary approaches such as read-across, molecular dynamics, and protein modeling are proposed to accelerate the development of toxicity information on PFAS, as well as the need for precaution to protect human health given the known effects of certain PFAS compounds.

Summary: PFAS are a class of synthetic chemicals that have been produced for over 70 years, with increasing concerns due to their widespread environmental presence, extreme persistence, bioaccumulative nature, and evidence for health effects from environmentally relevant exposures. Multiple US states have found the USEPA Health Advisories insufficiently protective and have developed more stringent guidelines for PFOA, PFOS, and other PFAS, with additional state guidelines expected to be released. The variations in state guidelines for PFAS reflect differences in toxicity and exposure considerations, with the range not unexpected given the different scientific approaches and historical context.

While current detection methods heavily rely on chromatographic techniques coupled with mass spectrometry, alternative methods such as optical and electrochemical techniques have been extensively explored for simple, accessible, inexpensive, rapid, and sensitive detection of PFASs. These alternative methods aim to overcome the limitations of expensive instrumentation, professional operators, complicated sample pretreatment, and considerable analysis time associated with chromatographic techniques.
