Brain Organoids: Tiny Mirrors of Human Neurodevelopment and Neurological Disorders

Anuradha Yadav et al.

Summary: Brain organoids, as 3D model systems, have been developed to study human brain development and neurological disorders. Despite their limitations, researchers are hopeful that future advancements with brain organoids could provide insights into the generation of neuronal circuitry complexity and reproducible creation of different human brain regions.

Human soluble ACE2 improves the effect of remdesivir in SARS-CoV-2 infection

Vanessa Monteil et al.

Summary: The combination therapy of remdesivir with recombinant soluble ACE2 shows significant therapeutic effects against SARS-CoV-2 in both cell and kidney organoid models. By targeting two different modalities of the virus's life cycle, this approach improves therapeutic windows. The study lays the groundwork for investigating combinatorial regimens in future COVID-19 clinical trials.

A host cell long noncoding RNA NR_033736 regulates type I interferon-mediated gene transcription and modulates intestinal epithelial anti-Cryptosporidium defense

Juan Li et al.

Summary: Intestinal epithelial cells play a crucial role in gastrointestinal mucosal immunity by generating barriers to protect against microbial infections. They quickly initiate innate immune reactions, including the production of antimicrobial molecules and release of chemokines and cytokines upon infection. Type I interferons, such as IFN-alpha/beta, activate immune effector cells and help in epithelial defense. Recent research on Cryptosporidium infection has highlighted the role of long noncoding RNAs (lncRNAs) in regulating type I interferon signaling in intestinal epithelial cells. The upregulation of specific lncRNA, NR_033736, acts as a negative feedback mechanism to fine-tune innate defense against microbial infections in the gastrointestinal tract.

Harmonization of Protocols for Multi-Species Organoid Platforms to Study the Intestinal Biology of Toxoplasma gondii and Other Protozoan Infections

David Holthaus et al.

Summary: Utilizing an in vitro platform with stem cell-derived intestinal organoids, models for human, mouse, pig, and chicken were established for studying mechanisms of infection by intestinal protozoan parasites. The generation, differentiation, and characterization of these organoids were optimized, leading to successful applications in comparative studies on parasite-host interactions and co-infections with relevant protozoans.

Effect of Listeria monocytogenes on intestinal stem cells in the co-culture model of small intestinal organoids

Jie Huang et al.

Summary: Infection with Listeria monocytogenes affects intestinal stem cells by damaging intestinal epithelium, reducing cell proliferation, and interfering with stem cell differentiation. It also increases mortality rates and impacts the expression of key genes in stem cell regulation. The study highlights the importance of using small intestinal organoids as an experimental model to study the interactions between intestinal epithelium and pathogens.
