Efficacy and safety of dupilumab for moderate-to-severe atopic dermatitis: A systematic review for the EAACI biologicals guidelines

Ioana Agache et al.

Summary: This review evaluated the efficacy, safety, and economic impact of dupilumab compared to standard care for uncontrolled moderate-to-severe atopic dermatitis. Dupilumab showed significant improvements in symptoms and quality of life for both adult and adolescent patients. Long-term safety data is needed for both children and adults, along with more efficacy data for the pediatric population.

Biofilm propensity ofStaphylococcus aureusskin isolates is associated with increased atopic dermatitis severity and barrier dysfunction in the MPAACH pediatric cohort

Tammy Gonzalez et al.

Summary: In the first US early-life cohort of children with atopic dermatitis, the presence of staphylococcal biofilms on skin lesions was found to be associated with increased disease severity, suggesting a pathogenic role forS aureus biofilms in atopic dermatitis development. Additionally, the balance between S epidermidis and S aureus, as well as their biofilm propensity, may have important implications for atopic dermatitis.

Systemic treatments in the management of atopic dermatitis: A systematic review and meta-analysis

Doreen Siegels et al.

Summary: This study critically evaluated systemic treatments for moderate-to-severe AD, with the most reliable and safe trial evidence found for the use of dupilumab in adults for up to 1 year. Limited evidence-based conclusions were drawn for other systemic treatments due to methodological restrictions. Therefore, head-to-head trials with novel systemic treatments are needed to clarify the future role of conventional therapies.

Microbial and transcriptional differences elucidate atopic dermatitis heterogeneity across skin sites

Noora Ottman et al.

Summary: This study found that different skin sites in atopic dermatitis (AD) patients have microbial heterogeneity, with distinct gene expression profiles between lesional and nonlesional skin. Keratinization dominates the transcriptomic signatures in the posterior thigh, while lipid metabolism is dominant in the upper back.

Increased occurrence of hand eczema in young children following the Danish hand hygiene recommendations during the COVID-19 pandemic

Anne B. Simonsen et al.

Summary: After implementing more intensive hand hygiene measures in Danish daycare centers, a high proportion of young children rapidly developed hand eczema. Prophylactic skin care could have potentially prevented some children from developing hand eczema.

Nasal DNA methylation differentiates severe from nonsevere asthma in African American children

Tao Zhu et al.

Summary: Significant differences in nasal epithelial DNAm were observed between nonsevere and severe asthma in African American children, a subset of which may be useful to predict disease severity. These CpG sites are subjected to the influences of environmental exposures and may regulate gene expression.

Summary: The common signature of AD across ages is T(H)2/T(H)22-skewed, but the specific expression of cytokines varies widely. Barrier abnormalities are present in all AD age groups, but significant decreases in filaggrin expression are only seen in adults.

Association between ambient air pollution and development and persistence of atopic and non-atopic eczema in a cohort of adults

Summary: The study found that increased exposure to residential ambient air pollutants was associated with an increased odds of eczema in males, but not in females. Additionally, there was a significant interaction between gender and ambient air pollution exposure and aeroallergen sensitization.

Electrical impedance spectroscopy for the characterization of skin barrier in atopic dermatitis

Arturo O. Rinaldi et al.

Summary: This study successfully established a method to directly assess the in vivo status of the epithelial barrier using electrical impedance spectroscopy, demonstrating its usefulness in patients with atopic dermatitis. Electrical impedance spectroscopy showed a significant negative correlation with transepidermal water loss, and a higher sensitivity in discriminating between lesional and non-lesional skin in patients with atopic dermatitis.

How does working in pandemic units affect the risk of occupational hand eczema in healthcare workers during the coronavirus disease-2019 (COVID-19) pandemic: A comparative analysis with nonpandemic units

Yasemin Erdem et al.

Summary: Healthcare workers in COVID-19 units are more prone to develop hand eczema (HE), with majority opting for increased use of moisturizing creams over topical corticosteroids for treating eczema. Non-COVID-19 unit workers have longer working years and more use of additional housework at home compared to COVID-19 unit workers. New strategies are needed for the prevention and management of HE among healthcare workers.

Does the epithelial barrier hypothesis explain the increase in allergy, autoimmunity and other chronic conditions?

Cezmi A. Akdis

Summary: The increase in allergic, autoimmune, and chronic diseases may be related to epithelial barrier-damaging agents associated with industrialization, urbanization, and modern life. These diseases are more prevalent in industrialized countries and are on the rise in developing countries, affecting over one billion people globally.
