X chromosome inactivation in female mammals results in cellular mosaicism and escape of some X-linked genes from inactivation, impacting the functional plasticity of female immune cells.

Summary: Objective evidence suggests that keratinocyte-fibroblast interactions are abnormal in systemic sclerosis (SSc). This study found that SSc epidermal equivalents (EEs) exhibited aberrant differentiation and enhanced expression of activation markers, replicating the abnormalities seen in SSc epidermis. The dysregulation of genes from the homeobox family and increased metabolic and oxidative stress in SSc EEs may contribute to chronic inflammation and dermal fibrosis.

Epidemiology of systemic sclerosis: a multi-database population-based study in Tuscany (Italy)

Summary: This study provided a comprehensive epidemiological characterization of Systemic Sclerosis in the Tuscany region, Italy, using a multi-database population-based approach. The prevalence of SSc was highest in individuals aged 65 and above, with females more predominant. Males with SSc had a more severe disease and lower survival rate, with significant differences in respiratory complications and metabolic comorbidities.

TGFβ promotes low IL10-producing ILC2 with profibrotic ability involved in skin fibrosis in systemic sclerosis

Summary: Our study demonstrated that in SSc patients, ILC2 numbers were increased in the skin and correlated with skin fibrosis severity. The dominance of natural ILC2 over inflammatory ILC2 in SSc skin was favored by the activity of TGF-beta. TGF-beta-stimulated ILC2 showed enhanced profibrotic activity by decreasing IL10 production and increasing myofibroblast differentiation, while therapeutic intervention in a mouse model combining pirfenidone with IL10 administration effectively reduced skin infiltrating ILC2 and alleviated skin fibrosis.
