Differences in Performance Characteristics Among Four High-Throughput Assays for the Detection of Antibodies Against SARS-CoV-2 Using a Common Set o Patient Samples

David M. Manthei et al.

Summary: With the passage of time from symptom onset, samples from SARS-CoV-2 RT-PCR-positive patients showed increasing positivity. The sensitivity of EUROIMMUN, Roche, and Siemens assays reached 93.1% to 96.6%, 98.3%, and 96.6%, respectively, for patients with the latest sample collected 14 or more days after symptom onset, outperforming the DiaSorin assay at 87.7%. The specificity of Roche and Siemens assays was 100%, higher than DiaSorin and EUROIMMUN assays.

Commercial Serology Assays Predict Neutralization Activity against SARS-CoV-2

Raymond T. Suhandynata et al.

Summary: This study demonstrates that three clinical serology assays positively correlate with SARS-CoV-2 neutralization activity observed in patients with COVID-19. All patients confirmed to be SARS-CoV-2 positive by PCR develop neutralizing antibodies. The agreement between the Diazyme IgG assay alone and the neutralization assay is 50.6%, while combining the Diazyme IgG assay with either the Roche or Abbott platforms increased the agreement to 79.2% and 78.4%, respectively.

Characterization of100sequentialSARS-CoV-2convalescent plasma donations

Christof Jungbauer et al.

Summary: The study analyzed antibodies in 100 convalescent plasma units for COVID-19, finding that high-titer plasma units tended to come from donors with more severe disease, older age, and male sex. However, there were limitations in the correlation between ELISA and neutralization assays, and further cases are needed to confirm the functional significance of this finding.

Evaluation of High-Throughput SARS-CoV-2 Serological Assays in a Longitudinal Cohort of Patients with Mild COVID-19: Clinical Sensitivity, Specificity, and Association with Virus Neutralization Test

Antonin Bal et al.

Summary: The study found that the Wantai Total Ab assay targeting the receptor binding domain (RBD) within the S protein showed the best sensitivity at different stages of the disease. With the exception of the Euroimmun IgA assay, all other tests had clinical specificity greater than 95%. Some commercial serological assays demonstrated better agreement with the VNT compared to others, but they cannot replace the VNT for assessing functional antibody response.

Multicenter Evaluation of the Clinical Performance and the Neutralizing Antibody Activity Prediction Properties of 10 High-Throughput Serological Assays Used in Clinical Laboratories

C. Therrien et al.

Summary: As the second wave of the COVID-19 pandemic emerges, screening population immunity is crucial to track infected individuals. Accumulating data on immune responses and nAb production in SARS-CoV-2-infected individuals will lead to new applications for serological assays. This study evaluated the clinical performances, reproducibility, and nAb level correlations of 10 immunoassays, with spike-based immunoassays showing better correlates of nAb activity.

Duration and key determinants of infectious virus shedding in hospitalized patients with coronavirus disease-2019 (COVID-19)

Jeroen J. A. van Kampen et al.

Summary: The study found that the median duration of shedding infectious virus in COVID-19 patients is 8 days, dropping below 5% after 15.2 days post onset of symptoms. High viral loads are associated with infectious SARS-CoV-2 isolation, while high serum neutralizing antibody titres are associated with non-infectious SARS-CoV-2.

Summary: A small study found that antibody responses significantly increased in healthcare workers after receiving a single dose of the SARS-CoV-2 vaccine, including antibodies to spike protein subunits S1 and S2, as well as the receptor-binding domain.

A Cross-Sectional Study of SARS-CoV-2 Seroprevalence between Fall 2020 and February 2021 in Allegheny County, Western Pennsylvania, USA

Lingqing Xu et al.

Summary: This study found a 4.5-fold increase in SARS-CoV-2 seroprevalence from Fall 2020 to February 2021 in Allegheny County, Pennsylvania, due to increased incidence of both natural disease and vaccination. The neutralization titer was significantly correlated with RBD titer but not with N titer. In the February cohort, higher median income and white race were associated with serological findings consistent with vaccination.

Summary: This study evaluated the specificity and sensitivity of 6 SARS-CoV-2 antibody assays, showing most methods performed well in terms of specificity and sensitivity. Sensitivity of tests more than 21 days after symptom onset varied among different methods, with similar average days of seroconversion.
