(Trifluoromethylselenyl)methylchalcogenyl as Emerging Fluorinated Groups: Synthesis under Photoredox Catalysis and Determination of the Lipophilicity

Kevin Grollier et al.

Summary: This study presents a method for synthesizing molecules bearing (trifluoromethylselenyl)methylchalcogenyl groups, with good yields up to 88% and up to 98% in flow chemistry conditions. The mechanism of this key reaction was investigated, and the emerging groups were physicochemically characterized. Additionally, the reaction was extended to perfluoroalkylselenolation with yields up to 95%, and successfully applied to the functionalization of bioactive molecules.

Trifluoromethyl Nonaflate: A Practical Trifluoromethoxylating Reagent and its Application to the Regio- and Stereoselective Synthesis of Trifluoromethoxylated Alkenes

Zhichao Lu et al.

Summary: The trifluoromethoxy group has sparked interest in drug and agrochemical discovery teams. TFNf is a reactive and scalable trifluoromethoxylating reagent that can be easily prepared in large scale and recycled after usage. The synthetic potency of TFNf was demonstrated through selective hydro(halo)trifluoromethoxylation reactions and nucleophilic trifluoromethoxylation of alkyl triflates/bromides and primary/secondary alcohols.

Direct Trifluoromethoxylation without OCF3-Carrier through In Situ Generation of Fluorophosgene

Jeremy Saiter et al.

Summary: Trifluoromethoxylation has attracted great attention in recent years due to the high interest in the OCF3 group for pharmaceutical and agrochemical applications. However, the existing methods often require the use of toxic, expensive, unstable F3CO- transfer reagents, which can generate undesired side-products. To address this issue, researchers have proposed a strategy involving the in-situ generation of gaseous fluorophosgene from triphosgene, its conversion by fluoride into the OCF3 anion, and the direct use of the latter in nucleophilic substitutions.

Radical C-H Trifluoromethoxylation of (Hetero)arenes with Bis(trifluoromethyl)peroxide

Stefan Dix et al.

Summary: The introduction of bis(trifluoromethyl)peroxide as a practical trifluoromethoxylating reagent opens up a new and efficient method for preparing trifluoromethoxylated arenes. This reagent, easily accessible from inexpensive bulk chemicals, allows for the preparation of trifluoromethoxylated arenes in good yields under mild conditions directly from unactivated aromatics using visible light photoredox or TEMPO catalysis. Additionally, TEMPO catalysis enables the one-step synthesis of valuable pyridine derivatives, simplifying the synthetic process.
