Baicalein suppresses growth of non-small cell lung carcinoma by targeting MAP4K3

Jian Li et al.

Summary: This study identifies MAP4K3 protein kinase as an important regulator in the development and treatment of non-small cell lung carcinoma (NSCLC). Baicalein, a natural product, is shown to induce autophagy in NSCLC cells by degrading MAP4K3, providing a potential therapeutic target for NSCLC treatment.

Foeniculum vulgare seed extract induces apoptosis in lung cancer cells partly through the down-regulation of Bcl-2

Weiwei Ke et al.

Summary: Ethanol extract of Foeniculum vulgare seeds (EEFS) significantly reduces lung cancer cell growth by targeting Bcl-2 protein expression and inducing apoptosis in vitro and in vivo. This study provides evidence for the potential of EEFS as a therapeutic drug for lung cancer.

Dynamic Regulation of Expression of KRAS and Its Effectors Determines the Ability to Initiate Tumorigenesis in Pancreatic Acinar Cells

Mohamad Assi et al.

Summary: The study demonstrates that pancreatic acinar cells become less sensitive to tumorigenesis induced by oncogenic Kras mutations after birth, due to low expression of KRAS and its effectors. Pancreatitis induces the expression of KRAS and its effectors, requiring the activity of EGFR. Expression of C-RAF in adult pancreas is essential for pancreatic tumorigenesis.

A comprehensive phenotypic CRISPR-Cas9 screen of the ubiquitin pathway uncovers roles of ubiquitin ligases in mitosis

Frances V. Hundley et al.

Summary: By conducting chemical-genetic CRISPR-Cas9 screens, we identified interactions between certain E3s/DUBs genes and compounds, which clustered functionally. Some genes interacted with multiple compounds, while others interacted primarily with a single compound or related compounds.

Inhibitors of cullin-RING E3 ubiquitin ligase 4 with antitumor potential

Kenneth Wu et al.

Summary: This study identified two compounds that inhibit E3 CRL4 and show potential as anti-tumor agents by affecting CRL4-mediated ubiquitination and causing cytotoxicity in tumor cells. Additionally, the research discovered that tumor cells with low levels of cullin4 protein exhibit increased sensitivity to these compounds, suggesting a new hypothesis for the role of low E3 abundance in cancer vulnerability.

Immune Stimulating Antibody-Photosensitizer Conjugates via Fc-Mediated Dendritic Cell Phagocytosis and Phototriggered Immunogenic Cell Death for KRAS-Mutated Pancreatic Cancer Treatment

Dahye Kim et al.

Summary: The research focuses on the development of CTX-conjugated maleimide-polyethylene glycol-chlorin e6 (CMPC) to enhance antitumor efficacy in patients with KRAS-mutated pancreatic adenocarcinoma. Results showed that CMPC combined with laser treatment significantly increased cytotoxicity and induction of immunogenic cell death, as well as enhanced phagocytosis and maturation of dendritic cells. In vivo biodistribution and antitumor effects demonstrated tumor selectivity and ablation efficacy, while infiltrated immune cells in tumor tissues and draining lymph nodes indicated innate and adaptive cellular immune stimulation.

Muscone derivative ZM-32 inhibits breast tumor angiogenesis by suppressing HuR-mediated VEGF and MMP9 expression

Liu-qing Yang et al.

Summary: In this study, a new muscone derivative, ZM-32, was identified with anti-angiogenesis effects mediated via targeting HuR in breast cancer. ZM-32 effectively prevented the proliferation and migration of breast cancer cells, inhibited the growth and angiogenesis of tumors in vivo, and influenced mRNA stability in a HuR-dependent manner. This compound may be a potentially valuable lead compound for anti-cancer angiogenesis.
