Natural Selection on the Phase-Separation Properties of FUS during 160 My of Mammalian Evolution

Pouria Dasmeh et al.

Summary: The study reveals that phosphorylation sites within the PLD of FUS are subject to stabilizing selection during evolution, with an exception seen in primates where the number of such sites has increased through positive selection. This work provides evidence that natural selection has stabilized the liquid forming potential of FUS and minimized the propensity of cytotoxic liquid-to-solid phase transitions during 160 million years of mammalian evolution.

Antivirals that target the host IMPα/β1-virus interface

Alexander J. Martin et al.

Summary: Small molecule inhibitors targeting the IMP alpha/beta 1-dependent nuclear import have shown robust antiviral activity, especially against viruses like HIV-1, DENV, and ZIKV. Compounds such as ivermectin directly target this interface and have shown broad-spectrum activity against various viruses, including SARS-CoV-2. Rigorous RCTs are underway to evaluate the therapeutic potential of targeting the IMP alpha/beta 1-virus interface for antiviral development.

Liquid-Liquid Phase Separation of Tau Driven by Hydrophobic Interaction Facilitates Fibrillization of Tau

Yanxian Lin et al.

Summary: The liquid-liquid phase separation of tau protein can facilitate amyloid aggregation in some cases, while being independent of aggregation in others. The nature of the interaction driving tau condensation is a differentiating factor between aggregation-prone and aggregation-independent liquid-liquid phase separations.

Observation of an α-synuclein liquid droplet state and its maturation into Lewy body-like assemblies

Maarten C. Hardenberg et al.

Summary: The study demonstrates that alpha-synuclein undergoes liquid-liquid phase separation to form liquid droplets, which then convert into an amyloid-rich hydrogel with Lewy-body-like properties. However, this maturation process towards the amyloid state is delayed in the presence of model synaptic vesicles in vitro.

Triad of TDP43 control in neurodegeneration: autoregulation, localization and aggregation

Paraskevi Tziortzouda et al.

Summary: The abnormal expression and cytoplasmic aggregation of TDP43, along with its nuclear loss, are characteristic features of neurodegenerative diseases. Autoregulation, nucleocytoplasmic transport, and phase transition are three intrinsic mechanisms that control the levels and localization of TDP43, essential for cellular homeostasis and affected in pathology.

TIA1 potentiates tau phase separation and promotes generation of toxic oligomeric tau

Peter E. A. Ash et al.

Summary: Tau protein interacts with RNA and RNA binding protein TIA1 to drive phase separation at physiological concentrations, generating toxic tau oligomers. This pathway may be a crucial source for toxic tau oligomers in tau-related neurodegenerative diseases. The selective mixing of proteins like tau and TIA1 in phase-separated droplets provides a basis for coassortment of functionally relevant proteins, potentially promoting specific biological activities.

Combating deleterious phase transitions in neurodegenerative disease

April L. Darling et al.

Summary: Protein aggregation is a hallmark of neurodegenerative diseases, but the mechanisms behind pathogenic aggregation remain unclear. Recent studies show that certain pathological proteins linked to neurodegenerative diseases can undergo liquid-liquid phase separation under physiological conditions, potentially leading to toxicity. Therapeutic strategies targeting aberrant phase transitions may offer a way to mitigate the toxicity and aggregation seen in these diseases.
