A Mechanistic Probe into the Dual Inhibition of T. cruzi Glucokinase and Hexokinase in Chagas Disease Treatment - A Stone Killing Two Birds?

Summary: The study showed that compounds GLK2-003 and GLK2-004 can simultaneously inhibit TcGLK and TcHK, with approximately 33.3% homology in their glucose/inhibitor binding sites. The total binding free energies of both compounds were favorable in both proteins, with key residues playing a crucial role in binding and stabilization. Both compounds induced a similar pattern of perturbations in the secondary structure of TcGLK and TcHK.
