Approaches for Systemic Delivery of Dystrophin Antisense Peptide Nucleic Acid in the mdx Mouse Model

Summary: The study compared the systemic administration of a 20-mer splice switching antisense PNA oligomer through intravenous and subcutaneous routes in mdx mice, showing slower excretion with subcutaneous administration and accumulation in kidneys and liver with depot technology release. Formulation of PNA as a duplex hybridization complex increased solubility, but none of the alternative administration methods resulted in significantly improved antisense activity. Further improvements in formulation or delivery methods may be necessary for enhanced pharmacokinetics and tissue targeting.

Vitamin B12-peptide nucleic acids use the BtuB receptor to pass through the Escherichia coli outer membrane

Summary: By covalently linking vitamin B-12 with PNA, researchers were able to utilize the vitamin B-12 transport system to deliver PNA to E. coli cells. The conjugate follows the same route through the outer membrane as free vitamin B-12, with BtuB playing a crucial role in the transport process. Molecular dynamics simulations provided insights into the mechanism of conjugate permeation through BtuB, highlighting the potential for hijacking the vitamin B-12 system for efficient PNA delivery.

Extended Peptide Nucleic Acid Nucleobases Based on Isoorotic Acid for the Recognition of A-U Base Pairs in Double-Stranded RNA

Summary: Peptide nucleic acids (PNA) with extended isoorotamides containing nucleobases (I-o) were designed to bind A-U base pairs in double-stranded RNA. Isothermal titration calorimetry and UV thermal melting experiments showed improved affinity for A-U using the Io scaffold in PNA, with PNAs having four sequential Io extended nucleobases maintaining high binding affinity.

Conformational constraints of cyclopentane peptide nucleic acids facilitate tunable binding to DNA

By incorporating helix-promoting cyclopentanes into peptide nucleic acids (PNAs), the melting temperatures of PNA+DNA duplexes can be finely tuned. The cpPNA-9 sequence specifically binds to complementary DNA at high temperatures and functions as a PCR clamp, likely induced by cyclopentanes promoting a right-handed helix in the PNA for DNA binding.
