Summary: This study established an in vitro CRISPR/Cas9 system with a novel recyclable, bidirectional selection marker gene amdS for efficient genome editing in Aspergillus niger. The endogenous amdS was confirmed as a dominant marker and can be removed by counter-selection, providing a powerful tool for marker-free genetic manipulation in filamentous fungi industrial-specific strains.
