Trigonelline inhibits Nrf2 via EGFR signalling pathway and augments efficacy of Cisplatin and Etoposide in NSCLC cells

Chandrani Fouzder et al.

Summary: The study demonstrated that Trigonelline inhibits Nrf2 activation and its nuclear translocation by blocking the EGFR signaling pathway, enhancing the responsiveness of NSCLC cells to Cisplatin and Etoposide without visible cytotoxicity to normal lung epithelial cells. It showed strong synergism in combination with Cisplatin/Etoposide at lower concentrations than the IC50 values of these drugs, indicating its potential as a promising choice for lung cancer therapy.

Non-canonical Glutamate-Cysteine Ligase Activity Protects against Ferroptosis

Yun Pyo Kang et al.

Summary: Cysteine plays a crucial role in maintaining cellular redox homeostasis and protecting against ferroptosis. Deprivation of cysteine leads to the unexpected accumulation of g-glutamyl-peptides in non-small-cell lung cancer cell lines as a protective mechanism against ferroptosis.

ATDC binds to KEAP1 to drive NRF2-mediated tumorigenesis and chemoresistance in pancreatic cancer

Vinee Purohit et al.

Summary: The overexpression of ATDC in pancreatic cancer promotes tumor growth and metastasis by protecting cancer cells from ROS through stabilizing NRF2. This mechanism enhances chemoresistance and modulates redox balance in cancer cells.

The deubiquitinase USP11 regulates cell proliferation and ferroptotic cell death via stabilization of NRF2 USP11 deubiquitinates and stabilizes NRF2

Chunjie Meng et al.

Summary: USP11 plays a critical role in stabilizing NRF2 through deubiquitination, thereby promoting cell proliferation and resistance to stress-induced cell death. Experimental evidence supports the high expression of USP11 in non-small cell lung cancer and its positive correlation with NRF2 expression.

NRF2-Dependent Bioactivation of Mitomycin C as a Novel Strategy To Target KEAP1-NRF2 Pathway Activation in Human Cancer

Liam Baird et al.

Summary: Activating mutations in the KEAP1-NRF2 pathway are found in approximately 25% of lung tumors, leading to aggressive tumor growth, chemoresistance, and poor prognosis. The study identified mitomycin C as significantly more toxic in cells with aberrant Nrf2 activation, with NRF2 target genes responsible for the enhanced bioactivation. This suggests mitomycin C as a potential candidate for drug repositioning to target untreatable NRF2 activation in human tumors.
