Consensus statement on standards and guidelines for the molecular diagnostics of Alport syndrome: refining the ACMG criteria

Judy Savige et al.

Summary: The meeting expanded the indications for screening for pathogenic variants in the COL4A5, COL4A3 and COL4A4 genes, refined the ACMG criteria for variant assessment, and identified mutational hotspots in the collagen IV alpha 5, alpha 3 and alpha 4 chains.

Prevalence Estimates of Predicted Pathogenic COL4A3-COL4A5 Variants in a Population Sequencing Database and Their Implications for Alport Syndrome

Joel Gibson et al.

Summary: This study estimated the frequencies of predicted pathogenic COL4A3-COL4A5 variants in populations without known kidney disease. The COL4A5 variants were found in at least one in 2320 individuals, and the predicted pathogenic heterozygous COL4A3 and COL4A4 variants affected one in 106 of the population. The results suggest the population frequencies for Alport syndrome need to be adjusted for individual variant penetrance and other factors.
