Past, presence, and future of allergen immunotherapy vaccines

Yulia Dorofeeva et al.

Summary: Allergen-specific immunotherapy (AIT) is a specialized treatment for allergic patients, aiming to induce immune protection against allergens. While AIT offers long-lasting and cost-effective benefits, its broad application is hindered by technical challenges such as allergen preparation quality and the risk of side effects.

ARIA digital anamorphosis: Digital transformation of health and care in airway diseases from research to practice

Jean Bousquet et al.

Summary: Digital anamorphosis is used to define a distorted image of health and care that may be viewed correctly using digital tools and strategies. The MASK strategy for digital transformation in rhinitis is based on validated digital tools and strengthens the ARIA change management strategy for airway disease prevention and management. Solutions for practical steps of digital care enhancement are proposed through the use of the MASK digital tool and the CARAT online clinical framework.

Induction of IL-10-producing type 2 innate lymphoid cells by allergen immunotherapy is associated with clinical response

Korneliusz Golebski et al.

Summary: IL-10(+) ILC2s play a crucial role in modulating grass-pollen allergy, attenuating Th responses, and maintaining epithelial cell integrity. Patients with grass-pollen allergy have lower levels of IL-10(+) KLRG1(+) ILC2s, but this ability to produce IL-10 is restored after immunotherapy treatment. The mechanisms underlying this involve modifications in retinol metabolic pathway, cytokine-cytokine receptor interaction, and JAK-STAT signaling pathways in the ILCs.

Passive Prophylactic Administration with a Single Dose of Anti-Fel d 1 Monoclonal Antibodies REGN1908-1909 in Cat Allergen-induced Allergic Rhinitis A Randomized, Double-Blind, Placebo-controlled Clinical Trial

Mohamed H. Shamji et al.

Summary: Treatment with anti-Fel d 1 monoclonal antibodies (REGN1908-1909) improved nasal symptoms in cat-allergic patients by inhibiting allergen-IgE binding, modulating cytokine and chemokine concentrations, and suppressing allergic responses mediated by Fc epsilon RI, Fc epsilon RII, and T-helper cell type 2.

Peptide allergen-specific immunotherapy for allergic airway diseases-State of the art

David C. Wraith et al.

Summary: Allergen-specific immunotherapy is the only way to change the natural immune response of allergic diseases, and using peptides based on CD4(+) T cell epitopes instead of whole allergens may overcome the challenges of current treatment methods.

Altered chromatin landscape in circulating T follicular helper and regulatory cells following grass pollen subcutaneous and sublingual immunotherapy

Hanisah Sharif et al.

Summary: This study aimed to evaluate the role of circulating CXCR5(+)PD-1(+) T follicular helper (cT(FH)) and T follicular regulatory (T-FR) cells in grass pollen allergic patients following different immunotherapy treatments. The findings showed dysregulation of cT(FH) cells in the grass pollen allergic group compared to nonatopic healthy controls, as well as induction of T-FR and IL-101 cT(FH) cells following specific immunotherapy. Changes in chromatin landscape were observed in cT(FH) and T-FR cells post-immunotherapy.

Decrease in CD38+ TH2A cell frequencies following immunotherapy with house dust mite tablet correlates with humoral responses

Sonia Luce et al.

Summary: This study confirms the importance of monitoring TH2A cell frequencies in allergic individuals and extends the observation to perennial allergies to HDM. CD38 may help better identify the subset of TH2A cells down-regulated by AIT. The coordinated pathways observed between cellular and humoral responses in immunoreactive individuals highlight the adaptive responses during AIT.

One Hundred Ten Years of Allergen Immunotherapy: A Broad Look Into the Future

Oliver Pfaar et al.

Summary: Allergen immunotherapy is the only disease-modifying treatment option for type 1-mediated allergic diseases, but there is still a need for improvement in clinical efficacy and tolerability. Current innovative approaches include the development of biologicals, allergen-derived peptides, recombinant allergens, Toll-like receptor agonists, other adjuvants, and novel application routes for future AIT.

Erika Jensen-Jarolim et al.

Summary: Allergen immunotherapy (AIT) in Europe utilizes allergoid strategy to destroy or mask IgE epitopes and uses adjuvants to precipitate allergens at the injection site for effectiveness and safety, while subcutaneous AIT in the United States mainly uses nonadjuvanted extracts without allergoids.
