Posttranslational chemical installation of azoles into translated peptides

Haruka Tsutsumi et al.

Summary: Azoles are five-membered heterocycles found in peptidic natural products and synthetic peptiodomimetics. Here the authors demonstrate a posttranslational chemical modification method for in vitro ribosomal synthesis of peptides with exotic azole groups at specific positions.

Ribosomal Synthesis of Macrocyclic Peptides with β2- and β2,3-Homo-Amino Acids for the Development of Natural Product-Like Combinatorial Libraries

Emel Adaligil et al.

Summary: The study reports the ribosomal synthesis of macrocyclic peptides containing one or more beta(2)-homo-amino acids, confirming their compatibility and translation efficiency. It also explores methods to prevent the incorporation of beta(2)-homo-amino acids through N-methylation, and the use of alpha,beta-disubstituted beta(2,3)-homo-amino acids to increase diversity in a single position of a macrocyclic peptide without significantly increasing the overall molecular weight.

Macrocyclic peptides exhibit antiviral effects against influenza virus HA and prevent pneumonia in animal models

Makoto Saito et al.

Summary: Researchers have developed a class of macrocyclic peptides named iHA, which can bind the influenza viral envelope protein hemagglutinin and inhibit virus infection by blocking adsorption and fusion. Particularly, iHA-100 shows powerful efficacy in inhibiting the growth of highly pathogenic influenza viruses and preventing severe pneumonia at later stages of infection in mouse and non-human primate cynomolgus macaque models, indicating its potential as a next-generation, mid-sized biomolecule.

Ribosomal Synthesis of Macrocyclic Peptides with Linear γ4- and β-Hydroxy-γ4-amino Acids

Emel Adaligil et al.

Summary: This study demonstrated the ribosomal elongation of linear and statines amino acids to expand the diversity of nonproteinogenic amino acids in combinatorial peptide libraries. The incorporation of statines into peptides could potentially confer important pharmacological properties, such as protease resistance and membrane permeability, and enhance the potential of future drug discovery.

A Selection of Macrocyclic Peptides That Bind STING From an mRNA-Display Library With Split Degenerate Codons

Chi-Wang Lin et al.

Summary: Recent advancements in mRNA display technology have allowed for the selection of peptides with non-natural amino acids, expanding the chemical diversity of macrocycles. A novel approach has been developed to increase amino-acid diversity without sacrificing natural amino acids, resulting in a high sequence complexity. This method has been successfully applied to select functional macrocycles binding to human STING, showing promise for future applications in immunoregulation.

In vivo modulation of ubiquitin chains by N-methylated non-proteinogenic cyclic peptides

Joseph M. Rogers et al.

Summary: A recent discovery of small, non-proteinogenic cyclic peptides capable of binding specific Ub chains has shown promising anti-cancer activity in vivo. This highlights the potential of non-proteinogenic cyclic peptide screening in rapidly identifying in vivo-active leads for targeting ubiquitin chains as an anti-cancer mechanism of action.

Summary: This article reviews the research progress of ribosomally-synthesized and post-translationally modified peptides (RiPPs) as of June 2020, including the discovery of new RiPP classes, deeper understanding of the mechanisms for the installation of post-translational modifications, and the mechanisms by which enzymes recognize leader peptides. Additionally, genome mining tools and strategies for RiPP engineering are also discussed in the review.
