Data File Standard for Flow Cytometry, VersionFCS3.2

Josef Spidlen et al.

Summary: FCS 3.2 is a revised flow cytometry data standard that aims to capture instrument conditions and measurement features more precisely, providing a uniform file format. The new version supports mixed data types to efficiently handle new types of data and use cases, but is incompatible with existing FCS file readers.

High-sensitivity flow cytometric assays: Considerations for design control and analytical validation for identification of Rare events

Ulrike Sommer et al.

Summary: This paper provides a detailed approach for designing and validating the sensitivity of rare event methods in flow cytometry, addressing the impact of panel design and optimization on the lower limit of quantification. It aims to offer best practices for the development, optimization, and analytical validation of flow cytometric assays designed to assess rare events. Note that clinical sensitivity validation, which addresses the positive and negative predictive value of test results, is not discussed in this paper.

Systematic comparison of high-throughput single-cell RNA-seq methods for immune cell profiling

Tracy M. Yamawaki et al.

Summary: Our systematic benchmarking of seven high-throughput single-cell RNA-seq methods with 21 libraries under identical conditions revealed that the 10x Genomics 5 'v1 and 3' v3 methods exhibit higher mRNA detection sensitivity and fewer dropout events. This facilitates the identification of differentially-expressed genes and improves the concordance of single-cell profiles to immune bulk RNA-seq signatures, providing useful metrics for selecting a high-throughput single-cell RNA-seq method for profiling complex immune-cell heterogeneity.

Best practices for optimization and validation of flow cytometry-based receptor occupancy assays

Ed Hilt et al.

Summary: Flow cytometry is well-suited for monitoring drug-to-target engagement through receptor occupancy assays (ROA), but there are challenges in the design and implementation, especially in cases of receptor modulation or low target-antigen expression levels. Best practices for designing, optimizing, and validating ROA are proposed to address these challenges.

New cytometry tools for immune monitoring during cancer immunotherapy

Shomyseh Sanjabi et al.

Summary: The success of cancer immunotherapy in the past decade has spurred a need to better understand the human immune system. Immune checkpoint blockade antibodies can reinvigorate T cells in cancer patients, but resistance mechanisms hinder full benefit for all patients. Early detection of biomarkers and resistance mechanisms is crucial for optimizing treatment outcomes.

High-throughput multiparametric imaging flow cytometry: toward diffraction-limited sub-cellular detection and monitoring of sub-cellular processes

Gregor Holzner et al.

Summary: This sheathless, microfluidic imaging flow cytometer with stroboscopic illumination achieves blur-free fluorescence detection at ultra-high analytical throughput, quantifying multi-parametric fluorescence and sub-cellular localization at 500 nm resolution. The effectiveness of the approach is demonstrated through analysis and localization of P-bodies and stress granules in yeast and human cells, with throughputs exceeding 60,000 and 400,000 cells/s. The utility of the imaging flow cytometer is highlighted in directly investigating phase-separated compartments within cellular environments and screening rare events at the sub-cellular level for various diagnostic applications.

Full Spectrum Flow Cytometry as a Powerful Technology for Cancer Immunotherapy Research

Diana L. Bonilla et al.

Summary: The focus is on understanding the reasons for checkpoint blockade failure and identifying immunological biomarkers that can predict treatment response, disease progression, or adverse effects. Flow cytometry, as a single-cell analytical tool, is used to monitor various types of cancer, but the conventional flow cytometry no longer meets the need to evaluate multiple features simultaneously, thus prompting the adoption of Full Spectrum Flow Cytometry (FSFC).
