Understanding disorder-to-order transitions in protein-RNA complexes using molecular dynamics simulations

Summary: Intrinsically disordered regions in proteins play significant roles in signaling, regulation, and binding multiple partners such as DNA, RNA, and proteins. Computational analysis is needed to understand their binding and recognition mechanisms. Molecular dynamics simulations of protein-RNA complexes show that disordered regions are important for RNA binding and undergo a transition from disordered to ordered conformation upon binding, contributing significantly to binding affinity. These disordered residues are often located at interfaces or as linkers between regions with similar movements, overlapping or flanking functionally important residues in recognizing protein-RNA complexes. This study provides insights into the role and recognition mechanism of disordered regions in protein-RNA complexes.

Mex3a was identified as a prognostic factor for overall survival in CRC patients, and knockdown of Mex3a inhibited tumor cell migration, invasion, and proliferation. Furthermore, hsa-miR-6887-3p was found to downregulate Mex3a expression and inhibit CRC tumorigenesis both in vitro and in vivo.
