Multi-omics Analysis of CRISPRi-Knockdowns Identifies Mechanisms that Buffer Decreases of Enzymes in E. coli Metabolism

Summary: By using CRISPRi to decrease enzyme levels in E. coli, this study found that metabolism can temporarily buffer fitness defects and identified several gene-specific buffering mechanisms. The research demonstrates that CRISPRi screens can reveal both global sources of metabolic robustness and local regulatory mechanisms that buffer decreases in specific enzymes.

In-situ generation of large numbers of genetic combinations for metabolic reprogramming via CRISPR-guided base editing

Summary: Reprogramming complex cellular metabolism requires simultaneous regulation of multigene expression. This study introduces a new method called BETTER, which utilizes CRISPR-guided base editors to generate a large number of genetic combinations in situ, simultaneously altering the expression of multiple genes. Application of this method in industrially important and model microorganisms demonstrates its potential for large-scale fine-tuning of multigene expression in various microorganisms.
